L.R. BUSH, D.W. HAACK, M. SHLAFER and B. LUCCHESI, Protective effects of ~-adrenergic blockade in isolated ischemic hearts, EuroPean J. Pharmacol. 67 (1980) 209--217. The protective effects of the fl-adrenergic blocking drugs, propranolol and atenolol, were tested in a model of global ischemia and assessed electron microscopically. Cat isolated hearts were perfused retrogradely with arterial blood drawn from donor cats. After a period of equilibration, isolated hearts were rendered globally ischemic for 1 h and subsequently reperfused for another hour. Hearts were then flushed with physiological salt solution followed by perfusion-fixation with cacodylate-buffered glutaraldehyde, containing ionic lanthanum. Lanthanum was included as a probe of myocardial membrane integrity. Left ventricular subendocardial samples were processed and examined electron microscopically. Nontreated hearts, which underwent normothermic ischemia and reperfusion, displayed extensive ultrastructural damage. Nonischemic and donor cat control myocardial tissue appeared normal in all respects. Hearts that received either propranolol or atenolol maintained their ultrastructural integrity, resembling controls. Ionic lanthanum proved to be reliable as a marker of membrane integrity and permeabil i ty, as nontreated hearts displayed intracellular deposi t ion.of the marker, indicating that deteriorations of membrane integrity occurred. The results suggest that ~-adrenergic blockade may be valuable in preserving myocardium subjected to ischemia and reperfusion.

Comparison between the effects of L-Carnitine and Acetyl-L-Carnitine on myocardial infarction size in ischemic heart
Introduction: In the present study, potential differences between the effects of L-carnitine (LC) and acetyl-Lcarnitine (ALC) on ischemia/reperfusion (I/R)-induced myocardial infarction size were investigated. Methods: Male Wistar rats were randomly divided into five groups and then anesthetized by sodium pentobarbital. Hearts of the animals were removed and quickly mounted on a Langendorff ...
متن کاملEffective protection by NHE-1 inhibition in ischemic and reperfused heart under preconditioning blockade.
We compared the protective effects of ischemic preconditioning (IPC) and the Na(+)/H(+) exchanger-1 (NHE-1) inhibitor cariporide in isolated rat hearts subjected to global ischemia (45 or 90 min) and 30-min reperfusion and determined the protective effects of cariporide under IPC blockade with the mitochondrial ATP-sensitive K(+) channel blocker 5-hydroxydecanoate (5-HD). With 45-min ischemia, ...
متن کاملRole of mineralocorticoid receptors in the mediation of acute effects of hydrocortisone in isolated ischemic rat heart
Introduction: Cardiac preconditioning is an important method to reduce the damage caused by prolonged ischemia. Previous studies have shown that corticosteroids have protective effects on the heart, however at high concentrations this effect may be reduced with unknown mechanisms. We hypothesize that the contradictory effects of hydrocortisone at high concentration may be mediated via minera...
متن کاملAlcohol consumption reduces ischemia-reperfusion injury by species-specific signaling in guinea pigs and rats.
We recently discovered that regular alcohol consumption reduces ischemia-reperfusion injury to the same degree as ischemic preconditioning in guinea pig hearts. Ischemic preconditioning, like this cardioprotective effect of alcohol, is mediated by adenosine signaling in guinea pigs. In rats, ischemic preconditioning may be mediated predominantly by α1-adrenergic signaling. To be certain that th...
متن کاملThe effect of asafoetida essential oil on myocardial ischemic-reperfusion injury in isolated rat hearts
Objective: Previous studies reported that asafetida from Ferula assa-foetida Linn. species and its essential oil (AEO) have antioxidant effects. In the present study, the effect of AEO was evaluated on ischemic-reperfusion injury in isolated rat hearts. Materials and Methods: Forty-eight male Wistar rats were divided into 6 groups: 1) control group, 2) vehicle group, 3-5) AEO groups and, 6) car...
